Extraplastidic site-specific factors mediate RNA editing in chloroplasts.
Single nucleotides in higher plant organellar mRNAs are subject to post-transcriptional alterations by RNA editing, typically resulting in changes of the encoded protein sequence. Although some information has been acquired on the general features of the editing processes in both plastids and plant mitochondria, the mechanisms and factors involved in the selective recognition of the nucleotide ...
متن کاملSite-specific photochemical RNA editing.
Photo-induced artificial RNA editing was demonstrated using photo-reactive oligonucleotides containing 3-cyanovinylcarbazole nucleoside. This non-enzymatic and sequence-specific methodology will make a major contribution to the elucidation of RNA functions including non-coding RNAs and to the development of drugs based on sequence-specific RNA editing.
متن کاملA site-specific factor interacts directly with its cognate RNA editing site in chloroplast transcripts.
RNA editing involves a variety of genetic systems and occurs by different mechanisms. In higher plant chloroplasts, specific sites of some transcripts are subject to C-to-U conversion. We have previously shown that site-specific trans-acting factors for psbE and petB mRNA editing bind corresponding cis-elements, which are located 5 nucleotides upstream from the editing site. Here we report that...
متن کاملMonomeric site-specific nucleases for genome editing.
Targeted manipulation of complex genomes often requires the introduction of a double-strand break at defined locations by site-specific DNA endonucleases. Here, we describe a monomeric nuclease domain derived from GIY-YIG homing endonucleases for genome-editing applications. Fusion of the GIY-YIG nuclease domain to three-member zinc-finger DNA binding domains generated chimeric GIY-zinc finger ...
متن کاملSite-specific uncoupling of photosynthetic phosphorylation in spinach chloroplasts.
In spinach chloroplasts which have been shaken with n-heptane, photophosphorylation coupled to the reduction of nicotinamide adenine dinucleotide phosphate is inhibited about 95% while the photoreduction of NADP+ (and ferricyanide) is stimulated as much as 50%. Photophosphorylation catalyzed by phenazine methosulfate, however, is inhibited only 50%. These data are interpreted to indicate that t...
